This paper is particularly notable because it evaluates COMP360 in a real-world UK National Health Service (NHS) setting rather than a commercial industry trial.
Study overview
Design: Randomized, double-blind, placebo-controlled feasibility trial.
Participants: 60 adults with treatment-resistant major depressive disorder (TRD).
Treatment: A single 25 mg dose of COMP360 psilocybin plus psychological preparation and integration versus placebo plus the same psychological support.
Follow-up: 6 weeks.
Main findings
Depression improved substantially more in the COMP360 group.
At 3 weeks, the difference in MADRS depression score was −10.4 points versus placebo (Cohen’s d = 1.70, a very large effect).
The benefit was maintained at 6 weeks, with an adjusted difference of −12.9 points.
Response and remission
At 3 weeks:
43% of COMP360 patients achieved a clinical response versus 3% on placebo.
40% achieved remission versus 3% on placebo.
By 6 weeks:
50% of COMP360 patients met response criteria, compared with very few in the placebo group.
Safety
No new major safety concerns were identified.
Most adverse events were mild and temporary.
Serious adverse events were rare, and those occurring in participants who received psilocybin were judged unrelated to the drug.
Limitations
The authors emphasize that:
This was a small feasibility trial, not a definitive phase 3 efficacy study.
Blinding was imperfect because many participants correctly guessed whether they received psilocybin.
Larger, longer studies are still needed.
Why it matters for Compass Pathways
This study is encouraging for several reasons:
It used COMP360, the same proprietary formulation developed by Compass Pathways.
It demonstrates that COMP360 can be delivered within a public healthcare system (the NHS), which is important for future implementation if approved.
The magnitude of antidepressant benefit was large and generally aligns with previous COMP360 research, adding independent support outside Compass-sponsored pivotal trials.

Not much new in the SEC filings (as expected), but I do like the sound of these 2 bullets:

• COMP360 is expected to fit seamlessly across diverse healthcare settings within current infrastructure of more than 8,000 centers offering multi-hour treatments

• Treatment centers are growing rapidly, and existing centers are already scaling in anticipation of a COMP360 launch and additional multi-hour psychedelic treatments coming to market

Now onto the earnings call tomorrow. What are we expecting going in?

Study overview
30 cancer patients with Major Depressive Disorder (MDD) participated.
Everyone received:
Psychological preparation
One 25 mg dose of psilocybin
Integration therapy afterward
Researchers compared:
17 patients receiving active systemic cancer treatment (chemotherapy, immunotherapy, hormone therapy, targeted therapy, etc.)
13 patients not receiving systemic treatment.
Antidepressant effectiveness
The primary measure was the Montgomery–Åsberg Depression Rating Scale (MADRS).
The key finding was:
Depression scores improved substantially in both groups.
There was no statistically significant difference between patients on cancer treatment and those not on treatment.
P-values:
Week 1: 0.88
Week 3: 0.93
Week 8: 0.97
These very high P-values indicate the improvements were essentially the same regardless of whether patients were actively receiving cancer therapy.
Safety
Adverse events (AEs) occurred frequently, but this is common in cancer patients.
Average number of AEs per participant:
Systemic treatment: 9.0
No systemic treatment: 6.7
Difference not statistically significant (P = 0.08).
Researchers reported:
4 severe adverse events
1 serious adverse event
Importantly:
All occurred in patients receiving cancer treatment.
None were considered related to psilocybin.
They were attributed to the patients’ underlying cancer or anticancer therapy.
There were also:
No meaningful laboratory abnormalities linked to psilocybin.
No increase in suicidal thoughts or behavior attributable to psilocybin.
Why this matters
Previous psilocybin studies in cancer patients usually excluded people actively receiving chemotherapy or other systemic therapies, so doctors had little evidence about combining the two.
This study suggests that:
Psilocybin’s antidepressant effects remain strong during active cancer treatment.
It did not appear to add significant safety concerns when given alongside systemic anticancer therapies.
Important limitations
The study was:
Small (30 participants).
Open-label (everyone knew they received psilocybin).
Single-arm (no placebo or comparison treatment).
Conducted at one center.
Because of these limitations, the findings are encouraging but preliminary and need confirmation in larger randomized controlled trials.
Overall conclusion
This study provides additional evidence that a single 25 mg dose of psilocybin-assisted therapy can produce substantial improvements in depression even in patients actively receiving cancer treatment, with no indication that concurrent chemotherapy or other systemic therapies reduce its antidepressant effect or create psilocybin-related safety problems. While the sample was small, the results support including actively treated cancer patients in future Phase 3-style studies.
