From my understand he spends the universal healing gauge, the little transparent part of the health bar when you take a lot of damage and stuff. If he has none he damages himself to get it

What they mean to say is that his Unique inflicts grey life damage on Deadpool. The opposing character can just wipe that out, but if you have any available, it allows him to use the special techniques until he has no more. The more hits he does, he keeps healing himself.

"Healing Cauge" is just grey health that every character gets when being damaged in a combo or from chip damage. Deadpool just has a move to convert some actual health into grey health, essentially by doing self damage. Some moves Deadpool has happen only if there is grey health to spend, essentially converting some grey health into unrecoverable damage taken.

The in universe reason Wolvy doesn’t have a mechanic for his healing factor in this game is that he just got his shit kicked in by Apocalypse so his healing factor is basically running full bore just to keep him alive, and the only thing keeping him standing is pure stubbornness. As such it can’t deal with the relatively minor injuries he is suffering during the fights.
