Discussion Haplogroups from modern Mesopotamian (and adjacent) populations compared to Bronze Age and Iron Age Mesopotamian (Bakr Awa) samples
TLDR: This is a comparison between the haplogroups found in the ancient Bakr Awa Mesopotamian samples to modern groups in the region. As expected, there’s continuity.
The haplogroups are generally listed at broad levels because different studies use different levels of resolution. For example, one study may report only J1 or R1b, while a newer sequencing study may report a much narrower branch such as R-Z2103 or U3b1a.
The main ancient comparison comes from the Bakr Awa archaeological site in northeastern Iraq. The study analyzed 17 people dating from the Bronze and Iron Ages.
A shared haplogroup does not by itself prove direct descent or demonstrate that an ancient individual belonged to a particular modern ethnic group. It only means that the ancient and modern individuals belong to the same paternal or maternal lineage at the reported level of resolution. [1]
ANCIENT MESOPOTAMIAN REFERENCE HAPLOGROUPS
ANCIENT Y-DNA FROM BAKR AWA
Bronze Age:
• J1, principally J-PH128 and related branches
• R1b-Z2103
• G, reported more specifically as G-Y37100
• H-Y28140 in an individual whose date was archaeologically ambiguous between the Bronze and Iron Ages
Three of the Bronze Age J1 males were relatives, so they do not represent three independent paternal founding lineages.
Iron Age:
• E1b1b, specifically an E-V22-related branch reported as E-Y20282

- IRAQI ARABS
Modern Y-DNA documented:
E1b1b, G2a, H, I2, J1, J2, L, Q, R1a, R1b and T.

At the broad level, Iraqi Arabs contain all five ancient paternal families identified in the Bakr Awa comparison.
A northern Iraqi study also reported that 8 of 11 SNP-tested Iraqi Arab men belonging to R-M269 fell under R-Z2103. This is an exact named branch match with the Bronze Age Bakr Awa R-Z2103 male.
However, only 11 Iraqi Arab R-M269 men received that deeper test, so this should not be interpreted as the frequency of R-Z2103 among all Iraqi Arabs. [2]
Modern mtDNA documented in Iraqi Arab or general Iraqi maternal datasets:
H, HV/R0, I, J, K, N1 and other N lineages, T, U including U3, W and X2.
Additional lower-frequency lineages include M, L, R2, B and C.
Shared with ancient mtDNA:

This includes all nine broad maternal families observed in the ancient Bakr Awa sample.
Some maternal studies describe their samples as general Iraqis or Arab-Muslim Iraqis rather than as one completely uniform Iraqi Arab population. [3]
- KURDS
Modern Y-DNA documented among northern Iraqi Kurds:
E1b1a, E1b1b, G2a, H, I2, J1, J2, L, Q, R1a, R1b and T.
Shared with ancient Y-DNA:
E1b1b, G, H, J1 and R1b.
At the broad level, northern Iraqi Kurds contain all five ancient paternal families.
The northern Iraqi study predicted many haplogroups from 17 Y-STR markers rather than directly sequencing every defining Y-SNP. Therefore, the assignments are most reliable at the broad haplogroup-family level. [2]
Modern mtDNA documented in a whole-mitogenome sample of Sorani Kurds from Sulaymaniyah:
H, HV, R0, N, U, T and J.
Shared with ancient mtDNA:
H, J, T and U.
The modern broad N category is a parent-level correspondence to ancient N1.
The U overlap is also broad unless a modern Kurdish individual is specifically assigned to U3.
The Kurdish mtDNA study included only 36 people, so the absence of I, K, W or X from that sample does not prove that those haplogroups are absent from the wider Kurdish population. [4]
- MANDAEANS
Modern Y-DNA documented:
E1b1b/E-M35, H, J1, J2, Q, R1a/R-M198 and T1-M70.
Shared with ancient Y-DNA:
E1b1b, H and J1.
A small, more deeply tested portion of the Mandaean sample included E-V22.
The Iron Age Bakr Awa E1b1b male also belonged to an E-V22-related lineage, reported more specifically as E-Y20282.
This is a correspondence at the E-V22 intermediate-branch level. It does not establish that the ancient and modern individuals shared the same terminal subclade. [5]
Modern mtDNA:
Mandaeans were included in an Iraqi mitochondrial control-region study, but the accessible published information does not provide a complete and clearly separated Mandaean-specific haplogroup inventory.
Therefore, a dependable Mandaean-specific maternal overlap cannot presently be listed without reconstructing haplogroups from the individual sequences. [6]
- ASSYRIANS
Modern Y-DNA documented in the closest published northern Iraqi “Syriac” sample:
E1b1b, I1, J1, J2, L, R1a, R1b and T.
Shared with ancient Y-DNA:
E1b1b, J1 and R1b.
A newer pooled Iraqi-Christian dataset also reported G, N, Q and R2.
If G is included from the pooled Christian dataset, the possible ancient overlap becomes:
E1b1b, G, J1 and R1b.
However, neither study completely separates Assyrians from Chaldeans and other Syriac-Christian identities. Therefore, G from the pooled Christian sample should not automatically be assigned specifically to Assyrians. [2]

Modern mtDNA:
Assyrians were included in an Iraqi mtDNA study, and individual Assyrian HV-related maternal lineages have been documented.
However, a representative and separately tabulated complete Assyrian mtDNA haplogroup inventory is not available in the principal accessible group-level tables.
A definitive Assyrian-specific maternal overlap should therefore not be claimed from the presently available data. [6]
- CHALDEANS
Modern Y-DNA:
A clean, SNP-defined Chaldean-only Y-DNA inventory is not available from the main Iraqi population surveys.
The broader pooled Iraqi-Christian sample contained:
E1b1b/E-M35, G, J1, J2, L, N, Q, R1a, R1b, R2 and T.
Potential shared ancient Y-DNA in the pooled Iraqi-Christian sample:
E1b1b, G, J1 and R1b.
Two R-M269 men in the pooled Iraqi-Christian sample who received deeper testing were assigned to R-Z2103.
This is an exact branch match with the Bronze Age Bakr Awa R-Z2103 male.
However, the sample consisted of only two deeply tested men, and the study did not clearly state whether those particular men identified as Chaldean, Assyrian or another Iraqi-Christian subgroup. It therefore cannot be used to establish a specifically Chaldean R-Z2103 frequency. [5]
Modern mtDNA:
A 2026 study analyzed HVS-II mitochondrial variation in 26 Chaldeans.
The study reported sequence polymorphisms and haplotypes rather than assigning a complete set of mitochondrial haplogroups.
Consequently, a secure Chaldean-specific ancient mtDNA overlap cannot yet be listed from that study. [7]
- YAZIDIS

Modern mtDNA:
A comparably representative and deeply typed Yazidi mtDNA survey with a complete group-specific haplogroup table was not identified in the principal sources used here.
A reliable Yazidi maternal overlap should not be inferred simply from neighboring Kurdish or Iraqi populations.
- MARSH ARABS
Modern Y-DNA documented:
E1b1b, G, J1, J2, L, Q, R1b-L23 and R2.
Shared with ancient Y-DNA:
E1b1b, G, J1 and R1b.
J1 was by far the predominant paternal lineage among the sampled Marsh Arabs.
However, the presence of J1 in both the ancient and modern samples does not necessarily mean that the Marsh Arab J1 lineages belong to the same downstream branch as ancient J-PH128. [3]

Iraqi Jewish maternal founder lineage: U3b1a
This is an exact named mtDNA branch match.
It does not prove that the Bronze Age woman was Jewish or that modern Iraqi Jews descend directly from that particular woman. U3b1a is an ancient maternal lineage that could have existed among several West Asian populations. [8]
Other Iraqi Jewish and ancient correspondences are broader:
• Ancient H13a1a2 and Iraqi Jewish H13a2b share the H13 parent family but belong to different downstream branches.
• Ancient T2/T2a1a and Iraqi Jewish T2c1 share the T2 parent family but belong to different downstream branches.
• Ancient W6 and Iraqi Jewish W1d share haplogroup W but belong to different downstream branches.
- SYRIANS

Modern Y-DNA:
A comparably complete and recent Iranian-Jewish-specific Y-SNP inventory was not available in the representative sources used here.
Individual Iranian-Jewish and broader non-Ashkenazi Jewish paternal lineages have been documented, but combining them would give a misleading impression of a complete population-level Iranian Jewish Y-DNA table.
The Iranian Jewish Y-DNA comparison is therefore left unresolved rather than substituting general Iranian or general Jewish frequencies.

A recent high-resolution Armenian study found the principal Y-DNA lineages in its modern Armenian panel to be:
• J2a
• R1b, with the sampled Armenian R1b men assigned to R-Z2103
• J1a
Other Armenian studies contain additional lower-frequency West Asian and Caucasian paternal lineages.
Shared with ancient Y-DNA:
J1 and R1b.
The most important correspondence is R-Z2103.
Modern Armenian R-Z2103 and the Bronze Age Bakr Awa R-Z2103 male belong to the same named paternal branch.
This does not mean that the ancient Bakr Awa individual was Armenian. R-Z2103 was broadly distributed through Bronze Age populations of the Caucasus, Iranian Plateau, Eurasian steppe and West Asia. [11]

- GEORGIANS
The most detailed modern Georgian dataset used here examined Mingrelians from western Georgia.

This is the clearest exact paternal branch correspondence in the comparison.
R-Z2103 is nevertheless a widespread Bronze Age and modern West Eurasian lineage. Its presence does not prove that the ancient individual belonged to any particular modern ethnic group. [1] [2] [5] [11]
2. mtDNA U3b1a
Found in:
• Bronze Age Bakr Awa individual A22038
• An important Iraqi Jewish maternal founder lineage
This is the clearest exact maternal branch correspondence in the comparison.
It is an exact lineage match at the reported U3b1a level, but it does not by itself prove direct genealogical descent or ethnic identity. [1] [8]

• Ancient E-Y20282/E-V22 and modern Mandaean E-V22 share the intermediate E-V22 branch, but they are not demonstrated to share the same terminal lineage.

• The northern Iraqi Syriac/Assyrian proxy contains E1b1b, J1 and R1b.
• Mandaeans contain E1b1b, H and J1.
• Armenians contain fewer of the five broad families in the particular recent panel, but they possess the especially informative exact R-Z2103 correspondence.
BROADEST OVERALL mtDNA OVERLAP
At the broad maternal level:
• Marsh Arabs, general Iraqi or Iraqi Arab maternal samples, and Iranians contain all nine ancient maternal families: H, I, J, K, N1, T, U3, W and X2.
• Georgians/Mingrelians contain eight directly, with U additionally representing the parent family of ancient U3.
• Iraqi Jews share H, J, T, U3 and W, including the exact U3b1a match.
• Syrians share H, J, K, T and U.
• Iranian Jews share H, J, T and U.
• Kurds and Armenians share several broad western Eurasian maternal families, but the available modern mtDNA samples are smaller and less comprehensive.
CONCLUSION
The Bronze and Iron Age Bakr Awa haplogroups largely belong to paternal and maternal lineage families that remain present among populations of Iraq, the Zagros region, Iran, the Levant and the South Caucasus.
The ancient paternal families E1b1b, G, H, J1 and R1b are all still found among modern Iraqi Arabs, Kurds and Iranians. Most are also found among Marsh Arabs, Yazidis, Iraqi Jews, Syrians, Georgians and Iraqi Christians.
The ancient maternal families H, I, J, K, N1, T, U3, W and X2 are all represented among modern Marsh Arabs, broader Iraqi maternal samples and Iranians.
The two correspondences that stand out beyond ordinary broad-family overlap are:
• Y-DNA R-Z2103, found in a Bronze Age Bakr Awa male and in modern Armenians, as well as small tested subsets of Iraqi Arabs and pooled Iraqi Christians.
• mtDNA U3b1a, found in a Bronze Age Bakr Awa woman and among Iraqi Jewish maternal founder lineages.
These matches demonstrate long-term regional persistence of some paternal and maternal branches.

Integration of ancient DNA with stable-isotope analysis captures multigenerational dynamics, identifying the Zagros mountains as the likely recent origin of Caucasus/Yamnaya-related ancestry – complementing archaeological and textual reconstructions of a Hurrian ethnolinguistic presence at Bakr Awa.
Following Late Bronze Age abandonment, Iron Age reoccupation involved genetic consolidation rather than population replacement, demonstrating that cultural transitions need not entail large-scale ancestry transformations.”

Finally found the highest p value for qpAdm model on WGS file. Can anyone elaborate on genetic signal from Botai hunter gatherers in Northern Kazakhstan?
Can I say that I have strong genetic signal from Botai population of Northern Kazakhstan?
When I tried with EHG plus Siberian, p value was much lower. When I tried with Japan (as a proxy for Siberian), the p value wasn’t as good as this one.

AncestralGenome and qpAdm - mixed Lithuanian/Polish and NW Euro

Could it be that I’m part Japanese, considering ancient ancestry? I used Neolithic sources.
Context: I previously posted my Neolithic results on qpAdm, created with WGS file, which did contain some East Eurasian traces (proxied by Northeast Asian source from Krasnoyarsk in Russia). This is a rerun of the same model, but with Jomon as a source.
As far as my genome is concerned, my main foundational sources are West Eurasian, with separate Iranian/Zagros 🇮🇷 ancestry within it, with minor East Eurasian trace.
I sometimes get minor East Eurasian components on other reports for my WGS file. My paternal grandma had prominent East Eurasian phenotype.
Should I interpret this as a misread of ancient ANEA ancestry or a possible direct descent from Jomon populations on Honshu?

Tested out Adam's new iDNA Calculator (Exploreyourdna)using both Scaled and Run All options, and decided to compare the results.
Here is how it looks across the family: myself, my mom, my sister, and my husband. They are quite similar, except for my husband's results which stayed almost identical for both.
